Sažetak
Disease resistance (R) genes that confer resistance to a diverse spectrum of pathogens have been isolated from a range of plant species. These genes share striking structural similarities indicating that certain signalling events are common to all or most plant defense systems. It hase been demonstrated that amplification of multiple DNA sequences from plant species by polymerase chain reaction (PCR) is possible with the use of degenerate oligonucleotide primers designed from short conserved motifs of cloned plant R genes. These sequences have been called resistance-gene analogs (RGAs). Genetic analyses have associated a number of these sequences to known gene loci that confer resistance to viruses, bacteria, fungi, or nematodes. Recent advances in the development of genetic maps in faba bean (Vicia faba L.) and chickpea (Cicer arietinum L.), have allowed the identification of genes and QTLs (Quantitative Trait Loci) for resistance to parasitic plants (Orobanche crenata Forsk.) and fungal diseases such as ascochyta blight, rust and fusarium wilt. The aim of this study is to clone and characterize NBS-LRR class resistance-gene candidate sequences from faba bean and chickpea by using degenerate oligonucleotide primers based on conserved motifs of known R genes. The development of suitable polymorphic markers based on the cloned RGA sequences will facilitate the assessment of their potential linkage relationships with mapped disease resistance genes in these two legumes, their use in marker-assisted selection (MAS) and may lead to molecular cloning of new R genes.
