Sažetak
Thymol, as hydrophobic phenol-type natural compound has a wide spectrum of antimicrobial activity with MICs below 0.05 % (m/V). Its antimicrobial efficacy could be increased by combining with EDTA, which enhances the permeability of the cell wall of Gram-negative bacteria. The purpose of this study was to determine antimicrobial activity of thymol (2.5, 25 and 50 mg/mL) in combination with Na2EDTA (0.1, 0.5 and 7 mM) against 13 species of clinical isolates of Gram-negative bacteria by cylinder plate assay (P. aeruginosa, P. vulgaris, P. mirabilis, E. coli, E. coli ESBL+, Kl. pneumoniae, Kl. oxytoca, Salmonella spp., Ewdardsiella spp., S. marcescens, Sh. boydii, Sh. flexnerii and V. mimicus). The most effective was combination of 7 mM of EDTA and 25 mg/mL of thymol which significantly increased the bactericidal activity in all tested species, determinate as inhibition zones of growth. The second most effective combination was 0.5 mM od EDTA with 25 mg/mL which were effective against 77% of bacterial species testes. P. aeruginosa was the most resistant against thymol alone, or combination of thymol and EDTA. In time-kill study we used E. coli ESBL+ and P. aeruginosa as models for predicting of bactericidal activity of thymol and combination of thymol and EDTA. During 30 minutes of time-kill study, thymol 50 mg/mL with 5mM EDTA significantly decreased the number of bacterial cells in comparison with thymol alone against P. aeruginosa. Using E. coli, the combination of 0.125 mg/mL of thymol and 5 mM od EDTA killed all bacteria in 30 minutes, while thymol alone killed 70% of inoculum. EDTA potentiates the bactericidal action of thymol, and theirs combination could be used as disinfectants.
