Sažetak
Background and Purpose: The products of phospholipid hydrolysis act as second messengers within the cell. In the present study we tried to determine whether phospholipase D (PLD) represented a transduction pathway for angiotensin II (ANG II) receptors in rat kidney cortical tubules. Materials and Methods: PLD activity was assessed by the accumulation of [H-3]phosphatidic acid (PtdOH) and [H-3]phosphatidylethanol (PtdEtOH) in tubules labeled with [H-3]alkyl-lyso-phosphatidylcholine and incubated in the presence of 1% ethanol. Results and Conclusion: ANG II elicited a time- and dose-dependent accumulation of [H-3]PtdOH and [H-3]PtdEtOH with an increase of 30 +/- 9% and 41 +/- 9% in controls, respectively (p < 0.01 and p < 0.02, respectively), during application of 10(-7) M for 2.5 min. PLD activation by ANG II was insensitive to PKC inhibitors GF 109203X (10(-5) M) and calphostin C (10(-6) M). ANG II activated PLD in freshly isolated renal cortical tubules.
