Sažetak
Flavonoids, natural substances with diverse biological activities, are universally present in vascular plants. Quercetagetin 3’ -methylether- 7-O-ß- D-glucopiranoside (MQ), isolated from the flowers of Centaurea rupestris L. is flavonoid known to possess antiphytoviral activity. In this study we induced different in vitro grown tissues: shoots, embriogenic callus (EC) and tissues consisting of callus and shoots, in order to test quantitative (determined spectrophotometrically ) and qualitative flavonoid content (determined by HPLC) in those tissues as opposed to naturally-growing C. rupestris tissues. The antiphytoviral activity of all tissue extracts against tomato bushy stunt virus (TBSV) was also investigated. Flowers and leaves from naturally-growing plants had the highest content of flavonoids, contained MQ and highly inhibited virus infection. Although the concentration of MQ was much higher in flowers than in leaves, extracts of leaves showed higher antiviral activity. Extract of EC exhibited antiviral activity and had the higher content of flavonoids than the extracts of other in vitro grown tissues, but lower than leaves and flowers of naturally-growing plants. MQ was detected in traces in EC as well as in all other in vitro grown tissues except in shoots grown on MS medium. Extracts of shoots from MS and &frac12 ; ; ; MS medium had the same content of flavonoids and the same inhibitory effect on virus infection, but MQ was detected in traces in shoots collected from &frac12 ; ; ; MS, while its concentration was significantly higher in shoots grown on MS. The fact that antiviral activity did not depend on the concentration of MQ in the tested tissues and even did not correlate with the flavonoid content, prompted us to search for other substances that might have contributed to the antiviral activity. However, naringenin and flavanone, known antimicrobial agents and possible precursors in hypothesized biosynthetic pathway of MQ, were not confirmed in any of the tested tissues. Coumaric acid, a direct precursor of caffeic acid and an early precursor of a number of flavonoids, was detected in all investigated tissues except in EC. Our results show that all the extracts of C. rupestris tissues exhibited antiviral activity which was not dependent on concentration of MQ, a known antiphytoviral agent.
