Nephrotoxicity of cisplatin (CP) in rats is manifested by two peaks of polyuria, e.g., 1 day (d1) and 5 days (d5), respectively, following a single i.p. injection. Previous studies have indicated organic cation transporters OCT1 and OCT2 in the cell basolateral membrane along the renal proximal tubule (PT) as mediators of the CP transport into the cell. A possible effect of CP on the expression and intracellular distribution of these renal transporters has not been investigated. In this work the rats were treated with CP and the expression of OCT1 and OCT2 was tested by Western blotting in isolated membranes and by immuncytochemistry in tissue cryosections d1 and d5 after the CP treatment. On d5, both transporters were strongly downregulated. On d1, OCT1 remained unaffected, while OCT2 in the PT S3-cells exhibited a clear redistribution from an intracellular compartment into the cell basolateral membranes. This phenomenon was absent in animals treated with colchicine, indicating a possible involvement of exocytosis in the OCT2 redistribution that was dependent on intact microtubules.
