Sažetak
During the process of oxytetracycline production S. rimosus excretes into the medium hydrolytic enzymes among which proteases, amylase and DNases were detected. A procedure for the concomitant isolation of alpha-amylase and DNase has been elaborated. It is based on fractional precipitation, gel filtration and ion exchange chromatography and gives both enzymes in electrophoretically homogeneous form. Dnase is shown to be a basic protein (pI 9.5-10.0) of Mr of 15 ooo D, stable at pH 5.0-10.0 and up to 40oC. The isolated enzyme is inhibited by EDTA and citrate and requires Mg2+ or Mn2+ for its activity. Analysis of degradation products of highly polymerized salmon sperm DNA and circular plasmid DNA suggests that S. rimosus DNase is an endo-nuclease.
