Cytotoxic T lymphocytes, CTL, and natural killer, NK, cells contain in their cytoplasmic granules a pore-forming protein, perforin, P, a cytolytic molecule that is a mediator in cell-mediated cytotoxicity, CMC. In unstimulated peripfheral blood lymphocytes, PBL, perforin is constitutively expressed in NK and gamma-delta T cells, high level of expression, and in CD11b+ subpopulation of CD8+ T cells, moderate expression. However P can be induced in the CD8+ CD11b+ subpopulation, as well as in some CD4+ T cells. The aim of the present investigation was to analyse the phenotype and prevalence of perforin-expressing immunocompetent cells in PBL of two patients with clinicaly definite and laboratory supported definite MS during and at the term pregnancy. Perforin and cell surface antigens, CD3, CD4, CD8, CD16 and CD56, were detected simultaneously by flow-cytometry. Upregulation of P expression in the middle of the pregnancy and downregulation at the term pregnancy was found in both MS patients. A significant increase of the percentage of P+ cells was found for total P+ cells and their T-cell subsets, CD4+ P+ and CD8+ P+, in the middle of pregnancy.
