Sažetak
Acute promyelocytic leukemia (APL) involves leukemic blasts expansion blocked at the promyelocytic stage and reciprocal translocation t(15 ; 17), which results in the fusion of promyelocytic leukemia (PML) and retinoic-acid receptor-α (RARα ) genes. NB-4 cell differentiation was induced by ATRA (0.5 µ M), with or without BMP-2, BMP-4, or BMP-6 treatment (1, 5 pg/ml), and confirmed by increase in granulocytic antigen CD11b expression on flow-cytometry. Promyelocytic NB-4 line expressed Alk-2, Alk-3, Alk-6, and RII BMP receptors, which made it responsive to BMP treatment. Combined treatment with ATRA and BMP-2, BMP-4, or BMP-6 significantly blocked the promyelocytic differentiation, evidenced as up-regulation of CD11b expression (more than 95% inhibition of CD11b expression by ATRA and BMP-2, BMP-4, or BMP-6 (1 pg/ml for both) compared to ATRA alone). Combination of ATRA and BMP-2, BMP-4, or BMP-6 had greatest influence on increase in Alk-6 expression, without significant changes in the expression of PML/RARA-oncogene.
