Sažetak
Cell adhesion molecules (CAMs) mediate cell-cell and cell-matrix interactions. Expression of E- and P-selectin, vascular adhesion molecule-1 (VCAM-1), and intracellular adhesion molecule-1 and -2 (ICAM-1, ICAM-2) on the surface of endothelial cells, as an answer to the pathophysiological stimulants, is considered as a main event in the development of the atherosclerosis. The influence of elevated glucose concentration and metformin on the E-selectin, VCAM-1, and ICAM-1 expression on human aortic endothelial cells (HAEC) was investigated. HAEC were cultured 4 h in a Medium 200 with low serum grow supplement, homocysteine (7 μ mol/l) and glucose in range of concentration (5.5, 8.0, 12.0 and 16.5 mM). Expression of CAMs were analysed by flow-cytometry using monoclonal antibodies. Controls were CAMs expression in the medium with a physiological glucose concentration (5.5 mM). The influence of metformin (10 μ g/ml) was analyzed in combination with each glucose concentration. Incubation of HAEC with 12 mM glucose increased expression of E-selectin for 16% and VCAM-1 for 48%. Expression of E-selectin was 4-fold higher and VCAM-1 was 16% when cells were grown with 16.5 mM glucose. Addition of metformin increased expression of E-selectin (approximately 7-fold) and decrease VCAM-1 expression (88 %) in a medium with 5.5 mM glucose. In conditions of elevated glucose concentration (12 mM), addition of metformin increased E-selectin (2-fold) and VCAM-1 (84%) expression. In conditions of significant high glucose concentration, expression of E-selectin was not changed, and expression of VCAM-1 was elevated (72%) compared to controls. Effect of glucose and metformin on ICAM-1 expression was not observed. In conclusion, high glucose concentration increased E-selectin and VCAM-1 expression in HAEC, while ICAM-1 expression wasn’ t changed. Metformin administration additionally increased expression of adhesion molecules which could contribute to development of macrovascular complications in diabetic patients.
