Sažetak
Osteophytes are newly formed osseous and cartilaginous tissue outgrowths forming mostly at the margin of osteoarthrotic joints. They are extremely variable and irregular in size and shape but it seems that they develope and growth by the same processes of intramembranous and endochondral ossification as bones in embryonal development. However, little is known about the factors and molecular mechanisms regulating their growth. Among growth factors and cytokines known to be expressed in osteophytes are IGFs, TGFb, PDGF and interleukins. Since BMPs family of growth factors have an important effects on skeleton during development and postnatal life in bone metabolism and regeneration, we decided to assay their presence in osteophytic tissues. BMP-3 & -7 have been determined in osteophytes dissected from 20 osteoarthrotic hip and knee joints removed from patients undergoing arthroplasty. They were recognized as small overhanging multiple lips at different stages of development. Tissue was fixed in 4% PFA, embedded undecalcified MMA and in parafine wax, cut serialy (4mm) on rotatory microtome Leica RM 2155 equipped with tungsten carbide knife. Tissue slices were mounted on APES coated glass slides, dried on 37º ; C prior to immunohistochemistry. Immunolocalisation was performed using BMP-3 and -7 monoclonal and polyclonal antibodies and immunoperoxidase coupled secondary antibodies with DAB as a detection system. The analysis showed that immunostaning for BMP-7 was positive in specific areas of all examined osteophytes. Chondrocytes in mature and hypetrophic zones stained positively. Highly vacuolated chondrocytes close to the mineralizing bone showed strong BMP-7 positivity. In general, matrix-bound staining for BMP-7 was observed in interteritorial cartilaginous matrix and osteoid. Strong matrix staining for BMP-3 was observed in the superficial fibrous tissue and less intense in matrix of the hypertrophic zone of cartilage. Also, positive matrix staining for BMP-3 was present in osteoid of underlaying trabecular bone. In conclusion, BMP-3 & -7 are expressed in human osteophytes showing a specific temporal and spatial distribution which may indicate that osteophytes represent mainly repair attempts in degenerating joints.
