Sažetak
Rheumatoid arthritis (RA) is an autoimmune disease characterized by proliferation of inflamed synovial membrane and bone destruction. In spondyloarthritis (SpA), inflammation and excessive reparative responses may lead to inappropriate cartilage and bone formation, causing joint ankylosis. The aim of our study was to test changes in the expression of selected BMPs in the peripheral blood mononuclear cells (PBMC) of patients with RA and SpA. Blood samples were collected from SpA patients, either with ankylosing spondylitis (AS ; n = 10) or psoriatic arthritis (PA ; n = 13), and RA patients (n = 10). cDNA was extracted from PBMCs, amplified by quantitative PCR using TaqMan assays (Applied Biosystems) for BMP-2 and BMP-6, and expressed as the relative amounts of RNA (mean ± SD) for target genes normalized to GPDH, using the standard curve method. The expression of BMP-2 was similar in all patients, whereas the expression of BMP-6 was significantly higher in RA than in both types of SpA PBMC samples (1.3 ± 0.7 in RA vs. 0.7 ± 0.4 in PA and 0.7 ± 0.3 in AS, ANOVA, p = 0.03). Our preliminary results indicate that RA, as a chronic inflammatory disease, has increased expression of BMP-6, which may reflect the systemic activation of inflammatory calls and lymphocytes, in contrast to SpA. Our further aim is to test the expression of other BMPs, their receptors, signaling molecules, and antagonists in a larger number of patients with different chronic joint diseases and correlate them with the disease parameters.
