Sažetak
The objective of this study was to determine metal and metallothionein (MT) content in indigenous mussels Mytilus galloprovincialis from the kaštela Bay, which is an industrial and urban centre of Dalmatia, Croatia. The mussels were sampled in January 1995, after spawning. The content of cadmium, lead, copper and zinc in wet digested composite samples of different mussel tissues (total edible part, gills and digestive gland)were determined by an electrochemical technique and electrothermal atomic absorption spectroscopy. After tissue homogenisation, MT fraction was isolated in the supernatant (30000xg, 4oC, 40 minutes). Purification was performed by thermal treatment only (70oC, 10 minutes). The MT content was determined by the modified Brdička reaction. The mass fractions of MT in the edible part, gills and the digestive gland of mussels have been reported as well as the 95% confidence interval for a mean. The results indicate that the digestive gland has the highest inducibility and protein extraction efficiency. Therefore, it is most appropriate mussel tissue for monitoring studies. The MT content per gram of respective mussel tissue decreases in the order: DIGESTIVE GLAND < EDIBLE PART <GILLS 3.5 mg 1.1 mg 0.5 mg Based on our previous results (Pavičić et al., 1993) on the MT content followed yearly in the digestive gland of mussels collected from a mussel farm in the Lim Channel, the mass fraction of MT (2.1 plus/minus 0.4)x10-3 could be considered as the baseline level, although more data are needed to define with more certainty the baseline for exposed MT levels. For survey studies it is essential to apply the transplanted mussels instead of indigenous, due to defined age.
