There are more than 100 human papillomavirus (HPV) types and approximately 40 infect the anogenital tract. In cervical precancerous lesions (LSIL and HSIL, low and high grade squamous intraepithelial lesions), a large variety of HPV types can be found, while in cervical cancer only high-risk (HR) or HR together with low-risk (LR) types. Many different techniques are currently being used for HPV detection and typing in clinical samples. The vaccination against HPV is becoming a reality in the nearest future and typing of HPV will be an important tool in the monitoring of vaccine efficiency. Therefore, the performance of different methods of HPV typing needs to be assessed. For this study, cervical samples of Croatian and Czech women diagnosed with HSIL and previously collected and HPV tested were used. Only HPV positive samples for types other than the most common HPV types 6/11, 16, 18, 31 and 33 or samples positive for multiple HPV types were selected. The detection and typing of HPV was done by consensus MY09/11 and type-specific (TS) PCR (Croatia) and by GP5+/6+ PCR and reverse-line blot assay (RLB) (Czech Republic). All samples were further analyzed by Line Probe Assay (LiPA) (Innogenetics) and Linear Array (LA) (Roche). The results of the performance of different techniques and comparison of interlaboratory results were statistically analyzed. All samples were successfully typed by LA (100%), while LiPA and RLB method did not reveal the type in all samples. In more than 50% samples, all 3 tests agree on at least one HPV type. In approximately 19% cases the results of all tests were in agreement even when multiple types were present. The discordant results were detected in 6% of samples. Larger series of the samples will be evaluated for poster presentation. Our preliminary data shows that LA system can more frequently detect low prevalent HPV types than LiPA and RLB.
