Sažetak
Only a few cytogenetical studies on Botrytis cinerea, an economically important phytopathogenic fungus of well known morphology and epidemiology, were conducted until today. The successful attempts in visualisation of mitotic chromosomes are rare, probably because of the small size of vegetative hifa and chromosomes, but also due to the specific process of mitosis (Shirane et al., 1988, 1989 ; Taga i Murata, 1994). Difficulties in observation of mitotic chromosomes are those concerning the nuclear membrane which remains intact throughout division of the nucleus, and, also, presence of the rigid cell wall which affects efficiency of chromosome staining. Shirane et al. (1988) overcome these physical barriers using germlings of conidia by bursting their tips with methanol-acetic acid treatment, which resulted in successful releasing of metaphase chromosomes. Our modification of Shirane's method significantly shortened time required to perform this chromosome-staining technique without reducing quality of result.
