PROBLEM: Granulysin is a cytolytic molecule expressed by human natural killer cells and activated cytotoxic T lymphocytes. Granulysin is lytic against a variety of tumor cells and microbes. Since little is known about the expression of granulysin at the maternal-fetal interface, we analysed its presence and distribution in the first trimester pregnancy decidua. MATERIAL AND METHODS: Decidual mononuclear cells were obtained by enzymatic digestion of decidual tissue and density gradient centrifugation. Granulysin expression and localization were analysed using cell permeabilisation method, flow cytometry and confocal microscopy. RESULTS: Decidua contains approximately 20% of granulysin positive cells (G+) and the majority of G is expressed in NK cells. Granulysin is stored mostly in cytoplasmic granules near the cell membrane and rarely colocalizes with perforin. Most of the decidual lymphocytes are both G+ and perforin+. CONCLUSION: Beside perforin, granulysin seems to be the second most abundant cytolytic molecule expressed at the maternal-fetal interface.
