Sažetak
NaHCO3 cotransport was studied in basolateral membrane vesicles (BMLV) isolated from rat renal cortex by the fluorescence quenching of pH sensitive dye, acridine orange (AO). The transporter was proved to operate with sodium as well as lithium, formerly considered as much more favorable substrate. N-ethylmaleimide (NEM) and ethacrynic acid inhibited activity of sodium-bicarbonate cotransport by 53% and 48%, respectively, while dicyclohexylcarbodiimide (DCCD) and amiloride completely lacked the inhibitory effect. Since the removal of one kidney stimulates prompt functional and anatomical changes in the remnant kidney, Na- HCO3 cotransport was studied in rats submitted to unilateral nephrectomy (ULN). No changes in sodium-bicarbonate cotransport were observed 2, 12 and 24 hours after unilateral nephrectomy in the kidney undergoing compensatory growth. However, after 48 hours, significant increase in cotransport activity was obtained (9.65± 0, 84 ∆ F/min vs. 7.48± 1, 13 ∆ F/min in control, p<0.02). This finding suggests that Na-HCO3 cotransport is not activated in the phase immediately following renal loss, and is probably not connected with the initiation of kidney growth.
