Sažetak
The direct macrophage spreading inhibition (MSI) test was performed on 10 guinea pigs 7, 28 and 63 days after sensitization with complete Freund's aduvant. A significant dose-dependent spreading inhibition was found in cultures with tuberculin. This inhibition was most pronounced on day 7 after sensitization. There was no spreading inhibition in 10 control animals injected with incomplete Freund's adjuvant. Similar spreading inhibition was obtained when cells from single animals, or pooled cells, were used. Both in the MSI and the macrophage migration inhibition (MMI) direct test the dose-response lines are parallel, but the MMI test requires less antigen to produce 50 percent inhibition. The dispersion of individual results for given dose of antigen is considerably greater in the MMI than in the MSI test. If compared with the effect of control culture supernatants (R), lymphokine containing supernatants (P) depleted of antigen caused a significant, dose-dependent macrophage spreading inhibition after 90 min of incubation. Following 12 hours of incubation the same P supernatants reversed its effect provoking stimulation of macrophage spreading. After proper incubation interval the P supernatant provoked both MSI and MMI.
