Sažetak
The aim of this study was to investigate the effect of worldwide pollutant pentachlorophenol (PCP) on the induction of micronuclei (MN) in haemocytes of freshwater snail Planorbarius corneus L. . Freshwater snails were exposed to different concentrations of PCP (10 mg/l; 100 mg/l, 450 mg/l; 600 mg/l) under laboratory conditions. The test solutions and water were changed daily during the 14 days of experiment. The samples of haemolymph were collected after 4, 7 and 14 days of exposure to PCP. The haemocytes were fixed in 1% glutaraldehyde and stained in 10 mg/ml Hoechst. The level of MN frequencies was expressed as the number of MN in 1000 cells (o/oo). The spontaneous level of MN frequencies ranged from 0.995 o/oo to 2.006 o/oo. Positive response (15.730 o/oo) was observed after 4 days of exposure to 600 mg/l PCP with the level of MN frequency 5 to 7 times higher than observed in other samples (from 2.006 o/oo to 3.413 o/oo). After 7 days of exposure the effect was dose-related, and the MN frequencies ranged from 7.928 o/oo (for 100 mg/l) and 8.964 o/oo (for 450 mg/l) to 13.972 o/oo (for 600 mg/l). After 14 days of exposure the frequency of MN ranged from 0.995 o/oo (for control) to 15.974 o/oo (for 450 mg/l) while in sample exposed to 600 mg/l the number of cells was too low to make the proper analysis. This test was also time-dependent for lower concentrations of PCP (100 and 450 mg/l). The frequency of MN significantly increased after 7 and 14 days of exposure with both concentrations.
