Sažetak
Centaurea rupestris L. is an endemic species which grows on the limited area of the Croatian coast with exclaves on Apennine and Balkan Peninsula. Flower extracts of Centaurea rupestris, as well as flavonoid quercetagetin 3’ -methylether- 7-O-ß- D-glucopyranoside isolated from the flowers, are known to have antibacterial, antifungal and antiphytoviral activity. In vitro clonal multiplication, rooting and acclimatization method was developed for this endemic species. The highest multiplication rate of 12 shoots per explant was achieved on Murashige and Skoog (MS) medium supplemented with 1 mM 6-benzylaminopurine and 2.9 mM gibberellic acid. Indole-3-butyric acid was the best root inducing auxin for C. rupestris. Rooting percentage achieved on half strength MS medium supplemented with 3 mM indole-3-butyric acid was 58%. Flavonoids were extracted by percolation in 45% and 90% ethanol and ether (1:1, w/v) from different plant tissues grown on MS medium supplemented with several combinations of plant growth regulators. The presence of quercetagetin 3’ -methylether-7-O-ß- D-glucopyranoside was confirmed by high performance liquid chromatography in all the tissues that developed shoots, but not in roots, transgenic roots or embriogenic callus. This result implies that synthesis of quercetagetin 3’ -methylether-7-O-ß-D-glucopyranoside is organ specific (flowers and shoots).
