While it is well known that polymorphonuclear neutrophils (PMNs) play essential role in host defence, mechanisms by which PMNs may act against cancer are not understood. The aim of our work was to monitor oxidative burst of PMNs in respect to the tumour development from the early phase to the advanced stages in experimental model of W256 carcinoma. W256 expanded on Wistar rats, denoted as WaWi, was transplanted into Spraque-Dawley rats and induced strong inflammatory response of PMNs. The antitumorous activity of PMNs lead to spontaneous regression of the tumour in 22/25 rats. Distraction of PMNs from WaWi by s.c. injection of Sephadex G-200 that causes local accumulation of PMNs at the site of injection remote from the WaWi allowed tumour development, thus the tumours regressed in only 8/25 animals. Inflammatory antitumorous activity of PMNs was not only associated with oxidative stress at the site of tumour transplantation, but also on the systemic level, as verified by the whole blood chemiluminescence, oxidative burst of PMNs, serum values of MDA and immunohistochemistry for the HNE-protein adducts at the site of tumour regression. The obtained results suggest that PMNs might play important role in defence against cancer in the early stages of the tumour development, while their oxidative burst is essential mechanism of the anticancer response which might even lead to the tumour regression.
