Sažetak
PUFA peroxidation plays an important role in various diseases such as atherosclerosis, (neuro)degenerative diseases, fibroproliferative diseases and cancer. Although these diseases have different pathology, they all include cellular alterations of PUFA metabolism and peroxidation, which are not well understood. Therefore, it is of importance to create simple experimental systems for studying lipid peroxidation on the molecular and cellular levels. For this purpose, we have developed a novel system based on yeast that is expressing a heterologus delta12 desaturase gene, which allows induced PUFA synthesis in a cell that normally does not produce these fatty acids. Although functional expression of the desaturase did not affect cellular morphology or growth in the absence of pro-oxidants, low levels of HNE-protein adducts were detected in a strain expressing the desaturase gene ; HNE-protein conjugate production was greatly enhanced in the presence of H_2O_2, with a concomitant decrease in cell viability, under conditions of desaturase expression. Furthermore, we observed difference in sensitivity to oxidative stress depending on the duration of the desaturase expression. Initially the mutants were more sensitive to oxidative stress than the wild type cells, but their increased sensitivity was reversible. This change in oxidative stress sensitivity was reflected in a change of the cellular oxidative and antioxidative parameters. Evidence suggests that the yeast expressing the heterologous desaturase gene adjusted to low level of endogenous lipid peroxidation and consequentially, became more resistant to exogenous oxidative stress.
