Sažetak
We have assessed the immunogenicity of Aujeszky's disease vaccine (ADV) containing a live attenuated virus (strain Bartha K/61 lacking nonessential glycoprotein gE) for porcine peripheral blood lymphocytes (PBL), retropharingeal (RPLNL) and bronchial lymph node lymphocytes (BLNL) using lymphocyte stimulation test. The cells were obtained from 6 noninfected 8-week-old pigs farrowed to the sows immunized with ADV. Using flow cytometric cell sorting we have phenotyped and quantified T and B cell subsets in the sampled tissues. The in vitro proliferation of RPLNL and BLNL to ADV (10^4 TCID_50 per well) was significantly higher (P < 0.05 ; < 0.01) than that of PHA- or PWM-stimulated (5 microg/ml) cells, respectively. However, the responses to Baypamun (5 microg/ml ; BPM, Bayer, Leverkusen, Germany), although lower than those of RPLNL and BLNL to ADV, were much stronger when proliferation of PBL was observed. cytolytic CD8a^+ T cells occur in equal proportions (30.7% in RPLN and 29.7% in BLN) in the lymph nodes tested. The resident CD4a^+ helper T cells were slightly elevated (35.7% and 42.3%, respectively) as comared to CD8a^+ cells. In both lymph nodes CD21^+ B cells were more frequent (35.2% in RPLN and 44.1% in BLN) than CD1^+ (20.5% and 17.8%, respectively) cells. It is likely that viral envelope glycoprotein antigens, such as gC, gB, gD, and gH, stimulated in vitro proliferation of rather regional lymph node lymphocytes than PBL. When BPM was applied with ADV the stimulation of RPLNL and BLNL was additionally enhanced. BPM alone seems to be a potent stimulator for porcine PBL.
