Sažetak
In the last decadde, many reports have underlined the pleiotropic nature of melatonin and other substances linked with pineal gland. They play a fundamental immunoregulatory role and are implicated as regulators of both normal and pathological growth. In this study we evaluated the effects of pharmacological pinealectomy (Px) on liver regeneration and on lymphatic morphostasis, since according to our previous data these two processes are closely interrelated. Px was made in AO rats by the evening injection of propranolol(PRO), given simultaneusly with or 2 days before partial hepatectomy (Hx).Hx rats in the control groups were treated with NaCl in the evening or with PRO and NaCl in the morning.The hepatic effects were evaluated by cell cycle analysis, bromodeoxyuridine index and by determination of class II expression in the liver. Simultaneusly the FACS analysis in the spleen was done. The data have shown that treatment with PRO in the evening, which induces Px, decreases the process of class II upregulation in regenerating liver and suppresses the mitogenesis in early phase of liver regeneration.These, inhibitory effects on liver growth were folowed by changes in lymphoid morphostasis. Melatonin, given in the evening abrogated the inhibitory effect of Px on hepatic mitogenesis, but did not improved the changes of class II expression in the liver.Simultaneusly, treatment with melatonin markedly increased the proportion of W3/25, OX-39 and MAC-1+ cells in the spleen.Similar, although less expressed changes were notice in experiments where Px preceded to partial hepatectomy. The data emphasize that pineal gland and its main hormone may play a regulatory role in processes that control the normal growth.
