Sažetak
We have previously shown that changes of neuro-endocrine homeostasis, induced by long acting somatostatin analog SMS-201-995, may abrogate the natural resistance of AO rats to the induction of EAE (inducibility of disease arose from 9 to 55%). In this work an attempt was made to correlate the changes noticed in clinical score of disease with the peripheral lymphatic status and functional reactivity of lymphocytes to specific and unspecific antigen challenge, as well as with the findings in CNS.For this purpose the phenotypic profile of cells in spleen and lymph nodes, blastic tranformation to Con A and myelin basic protein (MBP) and immunohistology in CNS were done. The time kinetic study has revealed that relaps and recovery phase of EAE in SMS-treated rats induces the characteristic changes in the proportion of W3/25, OX-8, OX-1 and OX-39 cells in spleen and variations in the intensity of blastic transformation to Con A. Before the relaps, the latter was significantly increased in comparison with saline treated group, indicating that SMS-induced changes in systemic immunity contribute to the altered susceptibility to EAE induction in resistant AO rats.
