Sažetak
It is widely recognized that the development of saturated genetic maps provides a valuable tool in many areas of plant genetics and breeding. The most efficient way to develop a genetic map of a species is the use of relatively inexpensive and highly polymorphic markers such as RAPDs or AFLPs in segregating F2 generation. However, the dominant nature of these markers hampers the complete classification of marker genotypes in F2 generation and thus the estimates of the recombination fractions between markers could be biased and unreliable. In order to analyse the accuracy of the faba bean map developed in the F2 generation, based predominantly on dominant markers (e.g. RAPDs), we have compared it to the map derived from F2:6 recombinant inbred lines (RILs) obtained by single seed descent method from the same F2 generation. Considering 81 common markers, the pairwise estimates of the recombination fractions (r) between markers were contrasted using heterogeneity test. Out of 431 estimates of r between markers, only 18 of them were significantly heterogeneous. A high degree of correspondence between maps indicate that the map obtained in F2 generation is sufficiently reliable for further analyses including mapping of quantitative trait loci (QTLs).
