Legionella pneumophila is a facultative intracellular pathogen that accounts for the majority of cases of Legionnaires' disease. The hallmark of Legionnaires' disease caused by L. pneumophila is the intracellular replication within phagocytes in the alveolar spaces. It has been shown in an in vitro model that Dot/Icm type IV secretion system is essential for intracellular replication. Here we examined intracellular replication of wt (wild type) strain of L. pneumophila AA100 and the dotA (defect in organelle trafficking) mutant in the lungs of A/J mice by transmission electron microscopy (TEM). After forming the phagosome L. pneumophilla AA100 replicated in the lungs of A/J mice, and the peak of replication was achieved at 24h after infection In contrast, the lungs of mice infected with the dotA mutant contained only one or two bacteria per phagosome. Our data show intracellular replication of L. pneumophilla AA100 in the ultrastructural level and importance of dotA gene in pulmonary infection of A/J mice.
