Sažetak
Legionella pneumophila is an intracellular pathogen that modulates biogenesis of its phagosome to svade endocytic vesicle traffic. The Z-egfoneZ/a-containing phagosome (LCP) does not acquire any Bidocytic markers and is remodeled by the endoplasmic reticulum (ER) during early stages. We show bat intracellular replication of L. pneumophila is inhibited in IFN-g activated bone marrow-derived nice macrophages and IFN-g activated human monocyte-derived macrophages (hMDM), in a dose-lependent manner. This inhibition of intracellular replication is associated with maturation of the LCP into a phagolysosome, as documented by the acquisition of Lamp-2, Cathepsin D, and the ly-osomal tracer Texas Red Ovalbumin, and with the failure of the LCPs to be remodeled by the RER. Ike conclude that IFN-g activated macrophages override the ability of L. pneumophila to evade endo-ytic fusion, and the LCP is processed through "default" endosomal lysosomal degradation pathway.
