Sažetak
The christal structure, sequence comparisons and site-directed mutagenesis of cholinesterases now make it possible to define distinct residues and discrete domains of acetylcholinesterase (EC 3.1.1.7) and butyrylcholinesterase (EC 3.1.1.8) which contribute to overall specificity for reversible ligands, carbamoylating agents and organophosphates. This review focuses on residues of enzymes directly implicated in catalysis and mechanisms of inhibition, reactivation, and aging. Structure and catalytic properties of human natural BChE variants with respect to their sensitivity to drugs are also discussed.
