Sažetak
The distribution of restriction fragment length polymorphism (RFLP) across the turkey and chicken ovalbumin locus has been mapped in genome DNA isolated from blood of 40 animals, representing four phenotype varieties. Using the same primers, polymerase chain reaction (PCR) was used for amplification of the ovalbumin gene of turkey and chicken genomic DNA. In addition to the size of amplified ovalbumin gene, the order of restriction fragments was determined by comparing a map of the EcoRI, HaeIII, PvuII and AvaI sites in turkey and chicken. The lengths of PCR products derived from 40 turkey genome DNA were 2.47 kb without significant differences. Two polymorphic restriction sites in a PCR products such as AvaI site at position 2150 bp and PvuII at 1718 bp allows genotyping of turkey and chicken. Our results suggest that the ovalbumin genetic loci are preserved in all four turkey phenotypes at the location of Hrvatsko zagorje. For the comparison the high polymorphic diversity of the ovalbumin loci between the two species, chicken and turkey, was observed.
