Sažetak
Bringing together sensitive SPS procedure for determination of heavy metals and multicomponent analysis by multiple linear regressions, original and simple methodology for simultaneous determination of metals in the mixture, as a new and unique scientific contribution for development of pharmacopoeial methodology was inaugurated. A very simple and fast one-step SPS procedure is based on sorption of heavy metals on PAN-resin (cation-exchanger Dowex 50W-X4 impregnated with PAN reagent) and direct absorbance measurement of colour product Me-PAN sorbed on solid carrier in a 1-mm cell. Optimal working conditions were: PAN-resin, 0.2 g, pH 8.0, time colour development, 30 min, and sample volume, 200 ml. This method provides determination of heavy metals at the ng mL-1 level important in pharmaceutical practice: zinc, lead, cadmium, cooper, nickel, mercury, manganese and chromium. A multicomponent analysis is checked by simultaneous determination of metals in synthetic mixtures with different composition and content of particular metal. Good agreement between experimental and theoretical amount of heavy metals is obtained from recovery test (78.3 - 110.0%). Proposed method enables determination of particular metal ion, limit of determination lower than 5 μ g mL-1, and eliminates the lead standard solution. Inaugurated procedure gives the new impulse to development of the novel pharmacopoeial procedure for heavy metals. The proposed method was applied to the determination of cooper in ascorbic acid, lead in glucose and zinc in insulin.
