Sažetak
Flavonoids are phytochemicals exhibitng a wide range of biological activities, among which are antioxidant activity and ability to modulate several enzymes or cell receptors. Some of them can induce point mutations with or without metabolic activation, and also can interfere with essential biochemical pathways. We have examined four, structurally related flavonoids (quercetin, fisetin, myricetin and luteolin) as well as one glycosilated flavanone (naringin) for thier ability to induce inhibition of mitochondrial dehydrogenases and their influence on cell membrane permeability as a indicator of primary damage of cells. Their antioxidant/prooxidant effects as well as thier influence on cellular level of glutathione in human laryngheal carcinoma (HEp) and thier cross-resistant subline (HEp CK2) cells were investigated. Quercetin increased cell membrane permeability in HEp cells and induced peroxidation as well as GSH in both cell lines. Peroxidation induced by quercetin was higher in Hep cells. Naringin induced decrease of glutathione, but did not show any effect on lipid peroxidation or membrane permeability in cross-resistant cell line. In HEp cell line, naringin has decreased spontaneous lipid peroxidation and prevented leakage of cell membrane and lipid peroxidation after treatment with hydrogen peroxide. Fisetin and luteolin caused small increase in lipid peroxidation and membrane leakage after treatment with hydrogen peroxide in HEp CK2 cell line. They increased lipid peroxidation in HEp cell line, but also, these two flavonoids showed synergistic effect with hydrogen peroxide on lipid peroxidation in HEp cell line.
