A significant number of horses immunized with tetanus toxoid (TTd) and toxin (TT) failed to respond adequately and produced low titers of antitetanus-toxin antibody. We tested whether an immune response modifier (IRM) BaypamunŽ (BPM), given parenterally, could enhance humoral and cellular immune responses in horses selected as tetanus hyperimmune serum donors. The BPM-treated or nontreated horses were repeatedly immunized with TTd and TT and sampled 9 weeks following the immunization regimens. Phenotypic and functional characteristics of their PBL were evaluate by either flow cytometry or lymphocyte stimulation test using common mitogens (PHA and PWM) and cell-mediated cytolysis against autologous lymphoblasts, respectively. Also, the quantification of serum anti-TTd/TT antibodies and detection of their respective immunoglobulin classes were performed. The PWM-induced proliferation in BPM + TTd/TT-treated horses (SI = 60.4) was about 50% stronger than that of TTd/TT-treated horses (SI = 27.7) before the plasmapheresis. The former horses had more CD2+ , CD8+ , ELA II+, and B-sub.+ cells, but less CD4+ cells than the later horses. The PBL from horses that received BPM exhibited stronger cytolytic activity in either DC (+ 37%) or ADCC (+ 57%) than did the cells from horses immunized with TTd/TT only. In BPM-treated horses the mean titers of antitoxin antibody were much higher (P < 0.05 or P < 0.01) at 5 and 9 weeks after onset of the immunization, respectively. These horses had higher serum levels of IgG (P < 0.05), and slightly, but not significantly elevated and IgG(T). The use of BPM, rather as an adjuvant than as an IRM, would be a useful and cost-effective approach for enhancing the specific immune responses of horses previously primed with TTd/TT.
