Sažetak
Dopamine (3,4-dihydroxyphenethylamine, DOP) was covalently bond to styrene.maleic anhydride copolymer (SMA) in order to prepare polymeric prodrug SMA-DOP as potentially form of dopamine. The coupling reaction was run under nitrogen atmosphere in dry N, N-dimethylformamide in the presence of sodium dithionite. The remainig anhydride rings were opened with water.
Shema
The resulting conjugate contained 42.8 % of drug. Release of dopamine from SMA-DOP conjugate was studied in vitro and the following rate constants were obtained: k = 2.27 x 10-3 min-1 for pH = 7.4 and k = 7.98 x 10-2 min-1 for pH = 9.2.
