Sažetak
Reactive oxygen species are generated during ischemia and are considered as the basic principle of tissue damage caused by oxidative stress, in particular if ischemia is followed by tissue reperfusion. Hemorrhage also makes preconditions for excessive production of reactive oxygen species and is considered as an additional factor of aggravation that can increase the risk of irreversible chain reaction of oxidative stress induced by ischemia. The most common "intermediary" substance of oxidative stress is hydrogen peroxide (H2O2) generated both in the chemically defined experimental conditions and in the living system, being produced during detoxification of the very reactive and toxic oxygen free radicals (hydroxyl radical and Superoxide anion). However, H2O2 also belongs to the reactive oxigen species because it is converted into hydroxyl radical in the presence of iron. Thus, determining plasma peroxides could be useful to estimate the dynamics and intensity of the onset of oxidative stress. A colorimetric assay was used to define whether ischemic and hemorrhagic stroke might differently reflect on the production of endogenous peroxide species in plasma. The values obtained during the first three days of hospitalization were compared between the two groups of stroke patients according to dynamic change, and with healthy controls. In patients with ischemic stroke, the levels of plasma peroxides were below normal throughout the period of observation. On the contrary, in patients with intracerebral hemorrhage plasma peroxides were above normal on day 1 and 3, but were transiently equal to normal values on day 2 of hospitalization. Throughout the study, plasma peroxides were significantly higher in patients with intracerebral hemorrhage than in thoses with ischemic stroke. Thus, measuring the levels of plasma peroxides might distinguish different systemic oxidative stress in patients with ischemic stroke or hemorrhagic stroke. Additional studies of the dynamic changes of plasma peroxides in stroke patients and analysis of the onset of diseases (including studies of oxidative stress markers and apoptosis in the brain) will help further evaluate the observed differences.
