Sažetak
In an attempt to find out whether dihydroergosine affects rat brain GABA-A receptor sites, we measured the affinity and the number of these sites by using 3H-muscimol, a potent GABA-A receptor agonist. Besides, we measured plasma GABA levels of these animals and tested the ability of dihydroergosine to inhibit GABA-A receptor binding in vitro. Kd and Bmax values, obtained by Scatchard analysis of the data, revealed no difference between the control and dihydroergosine treated rats. Two hours after dihydroergosine administration a significant increase of plasma GABA levels was noted in females but not in males. A more detailed study on the relationship between the brain and plasma GABA levels would be desirable.
