Sažetak
Bromocriptine, dihydroergotamine and dihydroergotoxine affected the synthesis of GABA in the substantia nigra and caudate nucleus, as evidenced by an increase in the aminooxyacetic acid (AOAA, inhibitor of GABA-T) induced accumulation of GABA. Moreover, a stimulation of GAD and GABA-T was obtained with bromocriptine. When administered in combination with allylglycine (inhibitor of GAD) the ergot drugs shortened the period between allylglycine injection and the onset of convulsions. The most potent in this regard was dihydroergotoxine. This drug also lowered the ED50 for picrotoxin. The results might suggest that the ergot drugs studied inhibit the GABA mediated transmission and consequently lead to a compensatory increase of turnover in GABAergic neurons.
