Sažetak
Na, K-ATPase is a P-type ATPase and responsible for active transport of Na and K across the plasma membrane in animal cells. The primary sequences of the alpha and the beta subunit from various tissues are known, but the topology of the alpha subunit including the location and number of transmembrane spanning regions is still uncertain, especially in the C-terminal half of the molecule. In the present study immunocytochemistry is applied on surface replicas of freeze-dried membranes with 2-D crystals of Na, K-ATPase and use to identify the extracellular and cytoplsmic side of the membrane crystals. The sidednes of the N- and C-termini as well as amino acids 815-828 and 889-903 of the alpha subunit are located with sequence-specific antibodies.
