Sažetak
The effect of temperature on binding of 125I-growth hormone on immobilized anti-growth hormone antibodies, raised in sheep, has been investigated. The antibodies were coupled to Sepharose 4B by CNBr method. At 22 oC the labeled antigen was bound to antibody in 180 minutes, while at 37 oC the equilibrium was reached after 60 minutes. At higher temperature (48 oC) the denaturation of the antibodies was noticed. The pH optimum of the binding reaction was found to be between pH 7.5 and 8.0. The greatest reduction in the ability to bind was noticed after addition of MgCl2, glycin.HCl pH 2.5, guanidium.HCl and KSCN, while urea, acetic acid, phosphate ions as well as higher amounts of NaCl did not show a significant effect on the reduction. Similar results were noticed for free antibody, raised in rabbit, although in this case the equilibrium of the reaction was reached after 20 hours at 22 oC.
