Sažetak
DNA polymerases alpha, -beta, -gamma and reverse transcriptase (RT) from human melanoma tissue were purified by successive column chromatography on DEAE-cellulose and phospho-cellulose. Polymerases-alpha and -gamma were further purified on hydroxylapatite column. Ionic, pH and primer-template specificity were determined for each enzyme. Purification-fold (measured under optimal conditions) for pol.-alpha was 231, for pol.-beta 174, for pol.-gamma 205 and for RT 1188. No terminal deoxynucleotidyl transferase activity was detected. M.W. of enzymes estimated by SDS-gel electrophoresis gave a value for pol.-alpha 160000, pol.-beta 38000, pol.-gamma 100000 and for RT 68000. Serological studies with purified RT were performed to estimate an antigenic resemblance to RT-ases from BEV, RD-114, SiSV, AML, RLV and to RT isolated from human myelofibrotic spleen.
