Sažetak
From 218 frozen human pituitaries adenohypophises were separated and homogenized in 0.05 M ammonium acetate buffer, pH 5.0. The homogenate was centrifuged, the pellet saved for prolactin isolation and to the supernatant was added cold ethanol (final concentration 12.5%). Precipitated material was used for isolation of human growth hormone (hGH). It was dissolved in 0.1 M NaOH, pH adjusted to 10.5 and filtered through glass wool column. The filtrate was acidified to pH 6.0 and precipitated with 2M ammonium acetate. The supernatant was adjusted to pH 4.9 and proteins were precipitated with 1.25 M ammonium acetate. This precipitate was dialysed against water and gel-filtered on Sephadex G-100 with 0.05M Tris-acetate pH 8.5. The preparation still contained all pituitary hormones. In order to obtain hGH as pure as possible, DEAE ion-exchange step was included. Different protocols were examined, i.e. different buffers, ion-exchange matrices and the purity of final product was examined.
