Sažetak
Efforts to develop compounds that inhibit viral DNA polymerases by interacting with templates may lead to the discovery of useful compounds exibiting a higher therapeutic index, i.e. a low cytotoxicity and a high antiviral activity. This approach, however, will not lead to the development of a specific inhibitor of the viral enzyme unless one finds a compound that binds specifically to 70 S RNA. The strategical approach to develop an inhibitor of this type is, at the present state of our knowledge, unthinkable. The second molecular approach, which has proved to be more useful and relatively specific in developing such inhibitors, is to design compounds that bind to the viral enzyme. One of these compounds is the polycytidylic acid containing 5-mercapto-substituted cytidylate units. The mode of action of this compound as an inhibitor of the viral DNA polymerase, and its biological implications are presented.
