Sažetak
PL-10.1 (GEPPPGKPADDAGLV) is a pharmacologically active pentadecapeptide. It has been of interest to develop a relevant analytical technique for the peptide detection in various biological samples. In this paper the immunological properties of the peptide will be described. The first step in any immunochemical technique is to rise a specific antibody. In the first set of experiments 7 structurally different immunogens, i.e. 5 different PL-10.1 - BSA conjugates, PL-10.1 - Poly Lys and PL-10.1 - MAP conjugate, were prepared and the rabbits were immunized. It was shown that conjugates of the PL-10.1 linked to the BSA via amino-groups developed a specific immunological response. When the peptide was linked to any of the examined carriers via its (C-terminal) carboxy groups, no significant immunological response was induced. The antisera were purified and IgG antibodies examined by ELISA and dot-blot techniques. Specific antibodies reacted to the C-terminal end of the peptide (concluded from the tests performed with 32 different peptide analogues). On the basis of these results, the second immunization experiment was performed. Rabbits were immunized with the intact peptide and its octapeptide-fragments (N-terminal octapeptide, middle section and the C-terminal octapeptide). Each of these peptides were linked to BSA via their N-terminal aminoacid, thus leaving a free C-terminus. The best and the most specific responses were obtained with the C-terminal PL-10.1 octapeptide-fragment linked to the BSA. These results indicate that the epitope and the immunogenic part of the PL-10.1 pentadecapeptide are located within the linear sequence of the last five C-terminal aminoacids.
