Viroids are small circular RNAs ranging from 246 to 401 nucleotides devoid of any coding capacity but still able to induce plant diseases. Citrus exocortis viroid (CEVd) is an economically important pathogen of citrus and a model for investigations of RNA structure-function relationships, molecular evolution and molecular pathogenesis. Gynura aurantiaca (Blume) DC. is an experimental systemic CEVd-host displaying characteristic syndrome and yielding high titer of the viroid. In order to establish a controlled in vitro system for investigations of CEVd host-pathogen interactions, we initiated culture of both CEVd-infected and healthy G. aurantiaca plants. Basal Murashige-Skoog medium (MS), MS supplemented with 6-benzylaminopurin (BAP) or kinetin were tested. All media supported plant growth. Healthy and CEVd-infected G. aurantiaca growing on BAP supplemented media showed the highest increase in total mass and decrease in the synthesis of cyanidin-tetraglucoside, plant’ s natural reddish-purple pigment. The stability of CEVd populations within different in vitro plant lines was investigated using RT-PCR followed by single strand conformation polymorphism (SSCP) analysis. CEVd populations isolated from plants grown on hormone supplemented media showed different SSCP-profiles than those grown either on basal MS medium or in the greenhouse. The significance of these conformation changes in view of the CEVd population fitness and its biological properties through differential media pressure remains to be determined.
