Sažetak
A methodological approach to the histochemical examination of the activity of placental isoenzyme of alkaline phosphatase in the eight placentas in endangered pregnancies, is presented. The activity of placental alkaline phosphatase was examined by testing three substrates by the simultaneous coupling method according to Burstone and by metal-precipitate method according to Gomori. The following substrates were tested: 1. naphthol AS-MX phosphate, 2. alpha-naphtylphosphate and 3. beta-glycerophosphate. As couplers, Variamine Blausalz B and Fast Blue BB were served. Left isomer of amino acid of phenylalanine was added to the incubation solution as inhibitor in growing concentrations. The shortest incubation time optimal for reading the results was achieved with the application of the metal-precipitate method according to Gomori, and lasted eight minutes. The thermostability of isoenzymes was determined by dipping specimen of placental tissue into host gelatine as embeding medium. The final product of the reaction deposited itself in the outer plasma- and basal membrane of the villous trophoblast respectively. The stromal elements of the chorionic villi, including vascular stroma, were always negative. Addition of phenylalanine to the incubation solution showed that placental alkaline phosphatase was sensitive to these amino acid as inhibitor. The character of the reaction inhibition corresponded in biochemical investigations to the established non-competitive type of inhibition.
