Sažetak
A method for gas chromatographic (GC) determination of chloramphenicol (CAP) residues in muscle tissue was developed and validated. Extraction of CAP residues was performed by the technique of matrix solid-phase dispersion (MSPD). The final extract was evaporated and the trimethylsilyl derivative of CAP was detected by GC-ECD and GC-MS. For quantification, the internal standard used was the meta isomer of CAP (m-CAP) for GC-ECD. The method was validated by determining accuracy (recovery of CAP from fortified muscle tissue samples and analysis of the certified reference material BCR-445), repeatability and detection and quantification limits. Mean recoveries were 93, 96 and 98% at the levels of 5, 10 and 15 μ g CAP/kg, respectively. The repeatability at the same levels was 13, 11 and 3%, respectively. The limits of detection and quantification with ECD were 1.6 μ g CAP/kg and 4.0 μ g CAP/kg, respectively.
