Sažetak
Extensive explorations in the last years has resulted in discovery and characterization of large number of different chromatin modifying complexes as promoter-associated transcriptional coactivators or corepressors. Protein complexes that modify chromatin structure can be classified into two groups, based on their modes of action. The first class represents chromatin modifiers which posses ATPase activity and remodel chromatin structure. This class is the best exemplified by yeast Swi/Snf complex which is highly conserved in eukaryotes, and homologous complexes have been purified from Drosophila and humans. Several other ATP-dependent chromatin remodeling complexes have also been identified in yeast and higher eukaryotes. The second class involves proteins with different enzymatic activities which bring about covalent modifications of the amino-terminal tails of nucleosomal histones and thereby alter chromatin structure. In this review we summarize current knowledge about factors and activities involved in the chromatin transition at the PHO5 promoter and discuss possible mechanisms of chromatin remodeling.
