Sažetak
T cell antigen receptor (TCR) and pre-TCR complexes are composed of heterodimers in association with dimers of signal transducing invariant subunits (CD3γ , -δ , -ε and ζ ). Inducible DNA deletion between two LoxP sites can be achieved in the mouse using Cre-ERT2, a fusion protein between a mutated ligand binding domain of the estrogen receptor (ER) and the Cre recombinase, the activity of which can be induced by Tamoxifen (Tam) but not natural ER ligands. To study the role of specific genes involved in activation and homeostasis of T cells in vivo we are developing a mouse strain with the inducible and T cell specific expression of Cre. The specificity and inducibility of Cre will be achieved by inserting a CreERT2 expression cassette under the transcriptional control of CD3ε chain promoter.
