Sažetak
1, 4-Dihydroisonicotinic acid derivatives (1, 4-DHIND) – a comparatively new class of physiologically active nitrogen heterocycles, possess a wide variety of pharmacological activities and are compounds with very low toxicity, but the mechanism of physiological activity of these compounds is not completely clear. Study was performed to obtain the data of some 1, 4-DHIND radical scavenging and antioxidant activities and their effects on cellular viability. Following compounds were tested: 2, 6-dimethyl-3, 5-diethoxycarbonyl-1, 4-dihydroisonicotinic acid sodium salt ; sodium 2-(2, 6-dimethyl-3, 5-diethoxycarbonyl-1, 4-dihydropyridine-4-carboxamido) glutarate (glutapyron) and sodium 2-(2, 6-dimethyl-3, 5-diethoxycarbonyl-1, 4-dihydropyridine-4-carboxamido) ethane-sulphate (tauropyrone). We have chosen a complex of methods for characterisation of radical scavenging and antioxidative properties of 1, 4-DHIND: N, N-Diphenyl-N’ -picrylhydrazyl (DPPH.) assay ; deoxyribose degradation assay ; ABTS radical cation scavenging assay ; metal ion catalysed peroxidation of phospholipids and ANTIOX-CAP assay. The influence of tested 1, 4-DHIND on cell viability was determined by MTT assay using HeLa cells. The obtained data showed that only 2, 6-dimethyl-3, 5-diethoxycarbonyl-1, 4-dihydroisonicotinic acid possessed radical scavenging and antioxidative activities which could be comparable with the standard antioxidant trolox. Taking together the presented data, 2, 6-dimethyl-3, 5-diethoxycarbonyl-1, 4-dihydroisonicotinic acid seems promising for further antioxidant activities in vivo investigation, while mechanisms of action of other compounds should be further investigated in different direction.
