Sažetak
Irinotecan is a topoisomerase-interactive agent. It has been widely used in treatment of metastatic colorectal cancer. The genotoxic effect of maximum single dose (700 mg/m2), recommanded monotherapy dose, 350, and recommanded combined therapy dose - 180 mg/m2 was studied on cultured V79 cells. Two and 24 hours treatment of V79 cells with these concentrations of irinotecan resulted in a concentration-dependent increase of micronucleated binucleate cells and slight, but not statistically significant, increase in the alkaline comet assay results. The frequency of MN cells was significantly higher in irinotecan treated cells then of control samples (non-drug treated cells). Mitomicyn C (0.5 micro g /mL) treated cells were used as a positive control in this study. Irinotecan treatment also caused alteration in the cell proliferation kinetics. This was evidenced by the increase in the frequency of mononucleate cells. The statistical significance of the results was determining using one-way ANOVA test. The results of the comet and micronucleus test show statistically significance in the tail length and tail moment as well as micronucleus frequency in drug-treated cells with respect to non-drug treated cells. Chromosome aberration analysis showed the induction of large DNA rearrangements, i.e. a lot of chromosome exchanges. Irinotecan treatment also caused a lot of fragmented nuclei in V79 cells.
