Sažetak
The aim of this study was to analyze the expression of the mannose receptor (MR) and possible natural ligands for its carbonhydrate recognition domain (CRD) in the first trimester human decidua. Immunohistology was used to prove MR+ cells in frozen tissue sections.The expression of the MR and CD86 or FITC-Dextran uptake were analyzed by macrophages untreated or pretreated by various concentrations of Tumor Associated Glycoprotein-72 (TAG-72), Mucin Is (MUCIs), Mucin III (MUCIII) and mannan or cultured with mentioned substances or IL-4. Intracellular labeling of IL-4 was performed in cord blood T cells cultured with isolated CD14+ cells pretreated with TAG-72, MUCIs, MUCIII, mannan or medium only. The MR was present by 75% of CD14+ cells.Its expression and FITC-Dextran uptake decreased following 18-hr culture in vitro. Secretory phase glycoproteins MUCIs and TAG-72, as well as positive controls (MUCIII or mannan) decreased binding of anti-MR monoclonal antibody (PAM-1 clone) and FITC-Dextran uptake by decidual macrophages on a dose dependent manner.They increased CD86 expression on CD14+ cells and their capability to increase IL-4 production in naïve T cells.IL-4 in turn increased MR expression by decidual macrophages. The presence of functionally active MR on CD14+ macrophages could support IL-4 cytokine production in the first trimester decidual tissue, rich in progesterone dependent glycoproteins.
